    Lignin extracted from Canna edulis Ker residues shows a strong inhibitory effect on α-glucosidase and a promoting effect on α-amylase. Protease activity inhibition may play a key role in disease processes, such as metastasis, tumor invasion and bacterial colonization. Hence, in the present study, the inhibitory mechanism of lignin on trypsin was examined, including the interaction type, thermodynamic parameters, structure, reaction site and molecular docking.

    The isolated lignin presented an inhibitory effect on trypsin activity with an IC

    value of 1.35 μmol L

    . This inhibition was a mixed linear type with a constant K

    of 3.92 μmol L

    . The lignin could bind with the key amino acid residue Ser195 on the active site of the trypsin molecule to inhibit its activity, and the phenolic hydroxyl group and -OH on the β-O-4 structure of the lignin molecule were the major groups bound with trypsin.

    Soluble solids concentration (SSC), dry matter concentration (DMC) and flesh firmness (FF) are important fruit quality parameters in stone fruits. This study investigated the ability of a commercial visible/near-infrared (NIR) spectrometer to determine SSC, DMC and FF in nectarine, peach, apricot and Japanese plum cultivars at harvest. The work was conducted in summer 2019/2020 on 14 stone fruit cultivars at Tatura, Australia. Two sub-samples of 100 fruit each were collected before and after commercial maturity (± 5 days) in order to maximize sample variability.

    Partial least square (PLS) regression models based on the second derivative of the absorbance in the 729-975 nm spectral region proved accurate for the prediction of SSC and DMC (R

     > 0.750). Only the model generated for SSC in ‘Golden May’ apricot was less precise compared to other cultivars. No visible/NIR models were accurate enough to predict FF in the cultivars under study (R

     < 0.750).

    This study demonstrated that the visible/NIR spectrometer was a reliable tool to monitor SSC and DMC in stone fruits at harvest but proved less useful for FF estimation. These results highlight the potential of visible/NIR spectrometry to evaluate stone fruit quality both in situ pre-harvest and in the laboratory after harvest. © 2020 Society of Chemical Industry.

The Neosebastes species, found in the western Pacific and south-east Indian Ocean, have received little attention from a research perspective. Samples of the bighead gurnard perch, Neosebastes pandus, collected from the lower west and south coasts of Western Australia, were used to undertake the first comprehensive investigation of the biological characteristics of a Neosebastes species. Opaque zones in sectioned sagittal otoliths were validated as forming annually. Female N. pandus grow to a significantly larger size, on average, than males and dominate the largest size classes, while males, growing to a smaller size, accumulate in the intermediate size classes. Although males were far less numerous than females in those age classes in which both sexes were found, males and females attain similar maximum ages >25 years. Neosebastes pandus spawns over a brief period between May (austral late autumn) and July (austral mid-autumn) when water temperature and day length are declining. The mean monthly gonadosomatic indices (GSI, IGS ) values of females during the spawning period are 37-50 times higher than those of males, which mature at an earlier age and smaller length compared to females. Histological examination of the ovaries of females indicate that their structure is consistent with “general” teleost ovarian anatomy and thus differs from Scorpaena, Helicolenus and Sebastes species whose ovaries are adapted for specialized reproductive modes. SN 52 nmr The reproductive strategy of N. pandus of maximizing the reproductive output of females, by this sex attaining a larger size than males and pair spawning (i.e., large disparity in IGS values) demonstrates the close lineage between the Neosebastes and other Scorpaeniformes, such as the Scorpaena, Helicolenus and Sebastes.

    Cholangiocarcinoma (CCA) is a very aggressive cancer showing the presence of high cancer stem cells (CSCs). Doublecortin-like kinase1 (DCLK1) has been demonstrated as a CSC marker in different gastroenterological solid tumors. Our aim was to evaluate in vitro the expression and the biological function of DCLK1 in intrahepatic CCA (iCCA) and perihilar CCA (pCCA).

    Specimens surgically resected of human CCA were enzymatically digested, submitted to immunosorting for specific CSC markers (LGR5 [leucine-rich repeat-containing G protein-coupled receptor], CD [clusters of differentiation] 90, EpCAM [epithelial cell adhesion molecule], CD133, and CD13), and primary cell cultures were prepared. DCLK1 expression was analyzed in CCA cell cultures by real-time quantitative PCR, western blot, and immunofluorescence. Functional studies have been performed by evaluating the effects of selective DCLK1 inhibitor (LRRK2-IN-1) on cell proliferation (MTS [3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenbition exerts anti-neoplastic effects in primary CCA cell cultures. Human DCLK1 serum might represent a serum biomarker for the early CCA diagnosis.
